چکیده

In various loading methods of whole tumor cells or their derivatives, cells fusion of dendritic cells (DCs) and tumor cells have some advantages for antigen presentation, and could up-regulate cytotoxic T cells (CTL) for multiple tumor antigens, including unknown antigens. However, the mechanisms of strong CTL productivity of fusion cells (FCs) are still unknown. Heat shock proteins (HSPs) are molecular chaperones that cross-present chaperoned antigenic peptides with MHC class I molecules. Herein, we focused on clarifying the potential of FCs for CTL production from the comparison of DCs pulsed with two kinds of tumor cell lysates, such as soluble tumor cell lysates or freeze-thawed tumor cell lysates. DCs, CD8+ T cells and tumor cells were prepared from ten patients with gastric cancer, and paired autologous tumor cells were used in all experiments. FCs of OK432-treated DCs and heat-stressed tumor cells (modified FCs) showed significant up-regulation of tumor-associated CEA and HER-2 antigen, and DC-related HLA-DR and co-stimulatory molecules (CD83 and CD86). FCs showed significantly higher IFN-gamma and CTL productivity of CD8+ T cells than DCs pulsed with soluble or freeze-thawed tumor cell lysates. IFN-gamma and CTL productivity of FCs was significantly increased by the heat stress of tumor cells. HSP70 mRNA levels and production of HSP70 protein of modified FCs increased significantly as compared with those of DCs pulsed with soluble or freeze-thawed tumor cell lysates. DCs pulsed with HSP70.PC extracted from modified FCs could enhance CTL productivity significantly more than that of DCs pulsed with HSP70.PC from soluble or freeze-thawed tumor cell lysate pulsed-DCs. The significant up-regulation of HSP70 mRNA and protein levels of modified FCs was related to the potential of CTL productivity. These results suggest that modified FCs possess stronger ability for MHC-restricted CTL production than DCs loaded with soluble or freeze-thawed tumor cell lysates.

Prokaryotic Expression of Influenza A virus Nucleoprotein Fused to Mycobacterial Heat Shock Protein70

Background and Aims: The novel approaches in influenza vaccination have targeted more conserved viral proteins such as nucleoprotein (NP) to provide cross protection against all serotypes of influenza A viruses. Influenza specific cytotoxic T lymphocytes (CTL) are able to lyse influenza-infected cells by recognition of NP, the major target molecule in virus for CTL responses. On the other hand,...

Generation of cytotoxic T lymphocytes by MHC class I ligands fused to heat shock cognate protein 70.

Immunization with gp96 and heat shock cognate protein 70 (hsc70) purified with in vivo bound naturally occurring peptides or bound to synthetic peptides by in vitro reconstitution has been shown to induce peptide-specific cytotoxic T lymphocytes (CTL). In addition, mycobacterial heat shock protein 70 covalently fused to ovalbumin (OVA)-derived fragments has been shown to generate MHC class I-re...

In Vivo Cytotoxic T Lymphocyte Elicitation by Mycobacterial Heat Shock Protein 70 Fusion Proteins Maps to a Discrete Domain and Is Cd4+ T Cell Independent

To gain insights into the mechanisms by which soluble heat shock protein (hsp) fusions can elicit CD8(+) cytotoxic T lymphocytes (CTLs) against the fusion partner, mycobacterial (Mycobacterium tuberculosis) hsp70 was dissected to ascertain whether a particular hsp domain is necessary, and knockout mice were used to determine whether the fusion protein's immunogenicity is dependent on CD4(+) T l...

Heat shock protein 70 is able to prevent heat shock-induced resistance of target cells to CTL.

Heat shock or transfection with heat shock protein 70 (Hsp70) genes has been shown to protect tumor cell lines against immune mechanisms of cytotoxicity. We have reported previously that heat shock confers resistance to CTL in the rat myeloma cell line Y3 that is Hsp70 defective. Evidence is now presented that Hsp70 is able to prevent the induction of the resistant phenotype. In Con A-stimulate...
